USES/FORMS/SOURCES
4-Aminodiphenyl is used in organic research (Lewis, 1993). It was previously used as a rubber antioxidant (Budavari, 1989; (Hathaway et al, 1991). It is no longer produced commercially in the US or in most other countries because of its carcinogenic activity (HSDB, 1995).
-CLINICAL EFFECTS
GENERAL CLINICAL EFFECTS
- 4-Aminodiphenyl is toxic by ingestion, inhalation, and skin absorption. Some references state that 4-aminodiphenyl may induce METHEMOGLOBINEMIA, while others do not mention this effect.
- 4-Aminodiphenyl exposure is associated with a high incidence of bladder cancer in humans. In experimental animals, it has produced bladder and liver tumors.
- 4-Aminodiphenyl is no longer produced on a commercial scale. Because of demonstrated carcinogenicity in experimental animals and humans, contact by all routes should be avoided.

TOXIC; inhalation, ingestion or skin contact with material may cause severe injury or death. Contact with molten substance may cause severe burns to skin and eyes. Avoid any skin contact. Effects of contact or inhalation may be delayed. Fire may produce irritating, corrosive and/or toxic gases. Runoff from fire control or dilution water may be corrosive and/or toxic and cause pollution.

SUMMARY
- FIRST AID - EMERGENCY RESPONSE GUIDEBOOK, GUIDE 153 (ERG, 2004)
Move victim to fresh air. Call 911 or emergency medical service. Give artificial respiration if victim is not breathing. Do not use mouth-to-mouth method if victim ingested or inhaled the substance; give artificial respiration with the aid of a pocket mask equipped with a one-way valve or other proper respiratory medical device. Administer oxygen if breathing is difficult. Remove and isolate contaminated clothing and shoes. In case of contact with substance, immediately flush skin or eyes with running water for at least 20 minutes. For minor skin contact, avoid spreading material on unaffected skin. Keep victim warm and quiet. Effects of exposure (inhalation, ingestion or skin contact) to substance may be delayed. Ensure that medical personnel are aware of the material(s) involved and take precautions to protect themselves.
FIRST AID EYE EXPOSURE - Immediately wash the eyes with large amounts of water, occasionally lifting the lower and upper lids. Get medical attention immediately. Contact lenses should not be worn when working with this chemical. DERMAL EXPOSURE - Immediately wash the contaminated skin with soap and water. If this chemical penetrates the clothing, immediately remove the clothing, wash the skin with soap and water, and get medical attention promptly. INHALATION EXPOSURE - Move the exposed person to fresh air at once. If breathing has stopped, perform artificial respiration. Keep the affected person warm and at rest. Get medical attention as soon as possible. ORAL EXPOSURE - If this chemical has been swallowed, get medical attention immediately. TARGET ORGAN: Bladder, and skin [bladder cancer] (National Institute for Occupational Safety and Health, 2007).
INHALATION EXPOSURE INHALATION: Move patient to fresh air. Monitor for respiratory distress. If cough or difficulty breathing develops, evaluate for respiratory tract irritation, bronchitis, or pneumonitis. Administer oxygen and assist ventilation as required. Treat bronchospasm with an inhaled beta2-adrenergic agonist. Consider systemic corticosteroids in patients with significant bronchospasm.
DERMAL EXPOSURE EYE EXPOSURE - DECONTAMINATION: Remove contact lenses and irrigate exposed eyes with copious amounts of room temperature 0.9% saline or water for at least 15 minutes. If irritation, pain, swelling, lacrimation, or photophobia persist after 15 minutes of irrigation, the patient should be seen in a healthcare facility.
ORAL EXPOSURE EMESIS: Ipecac-induced emesis is not recommended because of the potential for CNS depression and seizures. GASTRIC LAVAGE: Consider after ingestion of a potentially life-threatening amount of poison if it can be performed soon after ingestion (generally within 1 hour). Protect airway by placement in the head down left lateral decubitus position or by endotracheal intubation. Control any seizures first. ACTIVATED CHARCOAL: Administer charcoal as a slurry (240 mL water/30 g charcoal). Usual dose: 25 to 100 g in adults/adolescents, 25 to 50 g in children (1 to 12 years), and 1 g/kg in infants less than 1 year old. If methemoglobinemia is suspected in a patient exposed to 4-aminodiphenyl, refer to the Methemoglobinemia Meditext(R) Medical Management for more information. METHEMOGLOBINEMIA: Determine the methemoglobin concentration and evaluate the patient for clinical effects of methemoglobinemia (ie, dyspnea, headache, fatigue, CNS depression, tachycardia, metabolic acidosis). Treat patients with symptomatic methemoglobinemia with methylene blue (this usually occurs at methemoglobin concentrations above 20% to 30%, but may occur at lower methemoglobin concentrations in patients with anemia, or underlying pulmonary or cardiovascular disorders). Administer oxygen while preparing for methylene blue therapy. METHYLENE BLUE: INITIAL DOSE/ADULT OR CHILD: 1 mg/kg IV over 5 to 30 minutes; a repeat dose of up to 1 mg/kg may be given 1 hour after the first dose if methemoglobin levels remain greater than 30% or if signs and symptoms persist. NOTE: Methylene blue is available as follows: 50 mg/10 mL (5 mg/mL or 0.5% solution) single-dose ampules and 10 mg/1 mL (1% solution) vials. Additional doses may sometimes be required. Improvement is usually noted shortly after administration if diagnosis is correct. Consider other diagnoses or treatment options if no improvement has been observed after several doses. If intravenous access cannot be established, methylene blue may also be given by intraosseous infusion. Methylene blue should not be given by subcutaneous or intrathecal injection. NEONATES: DOSE: 0.3 to 1 mg/kg. Concomitant use of methylene blue with serotonergic drugs, including serotonin reuptake inhibitors (SRIs), selective serotonin reuptake inhibitors (SSRIs), serotonin and norepinephrine reuptake inhibitors (SNRIs), tricyclic antidepressants (TCAs), norepinephrine-dopamine reuptake inhibitors (NDRIs), triptans, and ergot alkaloids may increase the risk of potentially fatal serotonin syndrome.

Of 171 workers exposed to 4-aminodiphenyl for 1.5 to 19 years, 11% had bladder tumors. The tumors appeared 5 to 19 years after initial exposure (Hathaway et al, 1991). In a study of 503 exposed workers, there were 35 histologically confirmed bladder carcinomas and an additional 24 men with positive cytology (Hathaway et al, 1991).

When heated, vapors may form explosive mixtures with air: indoors, outdoors and sewers explosion hazards. Those substances designated with a "P" may polymerize explosively when heated or involved in a fire. Contact with metals may evolve flammable hydrogen gas. Containers may explode when heated. - 4-Biphenylamine has been used as a rubber antioxidant and dye intermidiate. It may have been released to the environment during it's production and use. According to some sources, it is no longer manufactured. It also may exist as an impurity in diphenylamine (HSDB, 2003).
ENVIRONMENTAL FATE AND KINETICS
If released into air, 4-aminodiphenyl would probably be associated with aerosols and particulate matter and be subject to gravitational settling. Degradation should occur due to direct photolysis, oxidation by ambient oxygen, and also by reaction with photochemically produced hydroxyl radicals (estimated half-life 6.9 hours in the vapor phase). No experimental data on these processes are available (HSDB , 1994).
SURFACE WATER If released in water, 4-aminodiphenyl would adsorb moderately to sediment and particulate matter in the water column. It adsorbs UV radiation and is sensitive to oxidation so it may photolyze in surface waters, be oxidized after binding to humic material and be oxidized by dissolved oxygen and alkoxy radicals (estimated half-life 14 days in sunlit eutrophic waters). No experimental data could be found on the degradation of 4-aminodiphenyl by these processes. There is evidence to suggest that 4-aminodiphenyl is biodegradable, but no data for natural waters were available (HSDB , 1994).
TERRESTRIAL If released in water, 4-aminodiphenyl would adsorb moderately to soil and be oxidized after binding to humic materials. Being sensitive to oxidation, it may be oxidized by other soil components such as cations. There is evidence to suggest that 4-aminodiphenyl is biodegradable, but no data in soil could be found (HSDB , 1994).
OTHER AIR: The estimated half-life of 4-aminodiphenyl in the air by reaction with photochemically produced hydroxyl radicals is 6.9 hours in the vapor phase (HSDB , 1994).
