USES/FORMS/SOURCES
USES p-Anisidine is used to prepare guaiacol via diazotization and hydrolysis ((HSDB, 1996); Lewis, 1993). p-Anisidine is used in the manufacture of azo dyes and as a chemical intermediate for Vat Red 29 (C.I. 71140), Disperse Orange 15 (C.I. 10350), and Azoic Coupling Component 13 ((HSDB, 1996); Lewis, 1993). When coupled with N-acetyl H acid, Acid Violet 12 is produced ((HSDB, 1996)). When the alkyl couples with 6,6'-ureyl-enebis-1-naphthol-3-sulfonic acid and is then coupled with 4- amino-m-toluene-sulfonic acid, Direct Red 24 is produced ((HSDB, 1996)).
o-Anisidine is derived from reducing o-nitroanisole with iron or tin and hydrochloric acid or by heating o-aminophenol with potassium methyl sulfate (Lewis, 1993). o-Anisidine is an intermediate in the production of Solvent Red 1 ((HSDB, 1996)). p-Anisidines are derived from reducing p-nitroanisole with iron filings and hydrochloric acid and by methylation of p-aminophenol (Lewis, 1993).

-CLINICAL EFFECTS
GENERAL CLINICAL EFFECTS
- p-Anisidine may be toxic by inhalation, ingestion, and dermal or eye contact.
- p-Anisidine is an irritant and has caused decreased nerve excitability in experimental animals. Symptoms of exposure may include headache, dizziness, contact dermatitis, sulfhemoglobinemia and methemoglobinemia, and the occurrence of Heinz bodies. In experimental animals, it has been nephrotoxic.
- o-Anisidine is a strong irritant and may be absorbed following dermal contact. Symptoms of inhalation exposure include headache, vertigo, increased sulfhemoglobin and methemoglobin levels, and frequent occurrence of Heinz bodies.
- In experimental animals, o-anisidine caused hematologic changes, anemia, and nephrotoxicity. Rats fed o-anisidine developed enlarged, black, and granular spleens.
- Anisidines have been carcinogenic in experimental animals.

TOXIC; inhalation, ingestion or skin contact with material may cause severe injury or death. Contact with molten substance may cause severe burns to skin and eyes. Avoid any skin contact. Effects of contact or inhalation may be delayed. Fire may produce irritating, corrosive and/or toxic gases. Runoff from fire control or dilution water may be corrosive and/or toxic and cause pollution.

SUMMARY

Move victim to fresh air. Call 911 or emergency medical service. Give artificial respiration if victim is not breathing. Do not use mouth-to-mouth method if victim ingested or inhaled the substance; give artificial respiration with the aid of a pocket mask equipped with a one-way valve or other proper respiratory medical device. Administer oxygen if breathing is difficult. Remove and isolate contaminated clothing and shoes. In case of contact with substance, immediately flush skin or eyes with running water for at least 20 minutes. For minor skin contact, avoid spreading material on unaffected skin. Keep victim warm and quiet. Effects of exposure (inhalation, ingestion or skin contact) to substance may be delayed. Ensure that medical personnel are aware of the material(s) involved and take precautions to protect themselves.
FIRST AID EYE EXPOSURE - Immediately wash the eyes with large amounts of water, occasionally lifting the lower and upper lids. Get medical attention immediately. Contact lenses should not be worn when working with this chemical. DERMAL EXPOSURE - Immediately wash the contaminated skin with soap and water. If this chemical penetrates the clothing, immediately remove the clothing, wash the skin with soap and water, and get medical attention promptly. INHALATION EXPOSURE - Move the exposed person to fresh air at once. If breathing has stopped, perform artificial respiration. Keep the affected person warm and at rest. Get medical attention as soon as possible. INGESTION EXPOSURE - If this chemical has been swallowed, get medical attention immediately. TARGET ORGANS - Blood, kidneys, liver, cardiovascular system, and central nervous system [in animals: tumors of the thyroid gland, bladder and kidneys] (National Institute for Occupational Safety and Health, 2007).
GENERAL Move victims of inhalation exposure from the toxic environment and administer 100% humidified supplemental oxygen with assisted ventilation as required. Exposed skin and eyes should be copiously flushed with water. Because of the potential for rapid onset of CNS depression or seizures with possible aspiration of gastric contents, EMESIS SHOULD NOT BE INDUCED. Cautious gastric lavage followed by administration of activated charcoal may be of benefit if the patient is seen soon after the exposure.
The following treatment information is for Methemoglobin Inducers, CNS Depressants, and Irritants (patients exposed to anisidines should be assessed for the presence of methemoglobinemia before treatment for this condition is undertaken). INHALATION EXPOSURE INHALATION: Move patient to fresh air. Monitor for respiratory distress. If cough or difficulty breathing develops, evaluate for respiratory tract irritation, bronchitis, or pneumonitis. Administer oxygen and assist ventilation as required. Treat bronchospasm with an inhaled beta2-adrenergic agonist. Consider systemic corticosteroids in patients with significant bronchospasm. Carefully observe patients with inhalation exposure for the development of any systemic signs or symptoms and administer symptomatic treatment as necessary.
DERMAL EXPOSURE DECONTAMINATION: Remove contaminated clothing and jewelry and place them in plastic bags. Wash exposed areas with soap and water for 10 to 15 minutes with gentle sponging to avoid skin breakdown. A physician may need to examine the area if irritation or pain persists (Burgess et al, 1999). Treat dermal irritation or burns with standard topical therapy. Patients developing dermal hypersensitivity reactions may require treatment with systemic or topical corticosteroids or antihistamines.
EYE EXPOSURE DECONTAMINATION: Remove contact lenses and irrigate exposed eyes with copious amounts of room temperature 0.9% saline or water for at least 15 minutes. If irritation, pain, swelling, lacrimation, or photophobia persist after 15 minutes of irrigation, the patient should be seen in a healthcare facility.
ORAL EXPOSURE Because of the potential for CNS depression or gastrointestinal tract irritation do not induce emesis. Significant esophageal or gastrointestinal tract irritation or burns may occur following ingestion. The possible benefit of early removal of some ingested material by cautious gastric lavage must be weighed against potential complications of bleeding or perforation. GASTRIC LAVAGE: Consider after ingestion of a potentially life-threatening amount of poison if it can be performed soon after ingestion (generally within 1 hour). Protect airway by placement in the head down left lateral decubitus position or by endotracheal intubation. Control any seizures first. ACTIVATED CHARCOAL: Administer charcoal as a slurry (240 mL water/30 g charcoal). Usual dose: 25 to 100 g in adults/adolescents, 25 to 50 g in children (1 to 12 years), and 1 g/kg in infants less than 1 year old. METHEMOGLOBINEMIA: Determine the methemoglobin concentration and evaluate the patient for clinical effects of methemoglobinemia (ie, dyspnea, headache, fatigue, CNS depression, tachycardia, metabolic acidosis). Treat patients with symptomatic methemoglobinemia with methylene blue (this usually occurs at methemoglobin concentrations above 20% to 30%, but may occur at lower methemoglobin concentrations in patients with anemia, or underlying pulmonary or cardiovascular disorders). Administer oxygen while preparing for methylene blue therapy. METHYLENE BLUE: INITIAL DOSE/ADULT OR CHILD: 1 mg/kg IV over 5 to 30 minutes; a repeat dose of up to 1 mg/kg may be given 1 hour after the first dose if methemoglobin levels remain greater than 30% or if signs and symptoms persist. NOTE: Methylene blue is available as follows: 50 mg/10 mL (5 mg/mL or 0.5% solution) single-dose ampules and 10 mg/1 mL (1% solution) vials. Additional doses may sometimes be required. Improvement is usually noted shortly after administration if diagnosis is correct. Consider other diagnoses or treatment options if no improvement has been observed after several doses. If intravenous access cannot be established, methylene blue may also be given by intraosseous infusion. Methylene blue should not be given by subcutaneous or intrathecal injection. NEONATES: DOSE: 0.3 to 1 mg/kg. Concomitant use of methylene blue with serotonergic drugs, including serotonin reuptake inhibitors (SRIs), selective serotonin reuptake inhibitors (SSRIs), serotonin and norepinephrine reuptake inhibitors (SNRIs), tricyclic antidepressants (TCAs), norepinephrine-dopamine reuptake inhibitors (NDRIs), triptans, and ergot alkaloids may increase the risk of potentially fatal serotonin syndrome. Carefully observe patients with ingestion exposure for the development of any systemic signs or symptoms and administer symptomatic treatment as necessary.

Workers exposed for 6 months to 0.4 ppm of anisidines in air for 3.5 hours each day experienced headaches and vertigo, increased sulfhemoglobin and methemoglobin levels, and continual occurrence of erythrocytic inclusion (Heinz) bodies (ACGIH, 1991).

Use precaution when choosing a container. Liquid anisidine will break down some forms of rubber, plastics, and coatings ((HSDB, 1996)). Containers of o-anisidine should be kept closed and protected from sunlight (ITI, 1988).
- ROOM/CABINET RECOMMENDATIONS
o-Anisidine should be stored in an environment not affected by heat, as it can contribute to the chemical's instability ((HSDB, 1996)). o-Anisidine should be stored at a site that is as close as possible to the area in which it is used ((HSDB, 1996)). Keep o-anisidine in only one section of the storage area and ensure that this area is properly labeled ((HSDB, 1996)). For p-anisidine, provide general ventilation or local exhaust ventilation and dilution ((HSDB, 1996)).

Toxic NOx fumes are emitted when o- and p-anisidine are heated to decomposition (Lewis, 1992). When anisidine decomposes, poisonous vapors and gases, such as carbon monoxide, may be released ((HSDB, 1996)).
REACTIVITY HAZARD

Build dikes to contain anisidine leaks and to keep the agent out of sewers and water sources (AAR, 1994). Cover o-anisidine spills with a combination of 9 parts sand to 1 part soda ash. Mix and then transfer all material into a paper carton stuffed full of ruffled paper. Burn the carton in an open furnace with extreme care or in a furnace with a scrubber and afterburner (ITI, 1988). Absorb liquids containing anisidine in vermiculite, dry sand, earth, or a similar material ((HSDB, 1996)).
o-Anisidine may be dissolved in a flammable solvent (alcohol, for example) and atomized in a combustion chamber that has an afterburner and scrubber (ITI, 1988). Waste management activities associated with material disposition are unique to individual situations. Proper waste characterization and decisions regarding waste management should be coordinated with the appropriate local, state, or federal authorities to ensure compliance with all applicable rules and regulations.

ENVIRONMENTAL FATE AND KINETICS
In the atmosphere, o-anisidine is thought to exist almost completely in the vapor phase. This expectation is based upon a vapor pressure of 0.014 mmHg at 25 degrees C ((HSDB, 1996)). p-Anisidine in the atmosphere also is expected to exist almost entirely in the vapor phase. This is based upon a vapor pressure of 0.006 mmHg at 25 degrees C ((HSDB, 1996)).
SURFACE WATER In water, o- and p-anisidine may bond securely with any humic matter that exists in suspended solids and sediments ((HSDB, 1996)). In natural sunlit water, o- and p-anisidine may react with photochemically generated oxidants ((HSDB, 1996)).
OTHER In natural sunlit water, the ordinary half-life for aromatic amines, such as o- and p-anisidines, reacting with photochemically generated hydroxyl radicals is 30 hours based upon an ambient hydroxyl radical concentration of 1 X 10(-9) mole/1. The half-life for both isomers reacting with alkylperoxy radicals is approximately 19 hours based upon the same hydroxyl radical concentration ((HSDB, 1996)). The reaction of o- and p-anisidine with photochemically generated hydroxyl radicals is thought to be the dominant fate process. Three hours is the estimated half-life for both isomers ((HSDB, 1996)).
BIODEGRADATION
- o-Anisidine ((HSDB, 1996)):
A substrate with a 60 ppm concentration of o-anisidine combined with 0.5 ml/L wastewater effluent inoculum produced no degradation after 42 days. When the o-anisidine concentration was changed to 60 mg/L and an activated sludge inoculum set at 1.5 ml/L, the degradation rate increased to 8% after 35 days. With a substrate concentration of 60 ppm and activated sludge inoculum at 10 ml/L, degradation reached 10% after 28 days. An aqueous mineral salts media inoculated with a mixed culture of soil microorganisms was combined with 20 mcg/L o-anisidine and resulted in the persistence of the o-anisidine after 64 days.
- p-Anisidine ((HSDB, 1996)) :
A substrate containing a p-anisidine concentration of 60 mg/L mixed with a 1.5 ml/L sludge inoculum produced approximately an 8% degradation in 14 days and 95% in 28 days. A substrate with a 60 ppm concentration combined with 10 ml/L sludge inoculum resulted in a greater than 90% degradation rate in 28 days. An aqueous mineral salts media inoculated with a mixed culture of soil microorganisms was mixed with 25 mcg/L p-anisidine and resulted in complete degradation of the p-anisidine in 64 days.
