GENERAL CLINICAL EFFECTS
- Pindone is poisonous in experimental animals by the ingestion, intravenous, and parenteral routes. It causes reduced blood clotting which leads to hemorrhaging (Lewis, 1992).
- Pindone is a vitamin K antagonist and causes inhibition of prothrombin formation (Hathaway et al, 1991).
- In rats, ingestion of a single large dose of pindone causes rapid death due to pulmonary and visceral congestion without hemorrhage which may not be related to vitamin K antagonism. Death in animals from chronic exposure is due to multiple internal hemorrhages (Hathaway et al, 1991).
- Symptoms resemble those from warfarin (Budavari, 1989).
Warfarin is used as a drug, whereas pindone is not. There has been less experience with pindone; however, clinical use of indandiones has resulted in diarrhea, pyrexia, renal tubular necrosis, paralysis of accommodation, and exfoliative dermatitis (ACGIH, 1991).
- The following clinical effects are for LONG-ACTING ANTICOAGULANTS:
In massive overdose, these agents have produced rapid and persistent hypoprothrombinemia and associated bleeding diathesis. Children with ingestions of "mouth full" amounts have developed signs of anticoagulation. Once coagulation defects occur, they may persist for 6 weeks to many months. Cardiopulmonary injury and neurologic toxicity have been reported with the indandiones in experimental animals, but not in humans.

Highly toxic, may be fatal if inhaled, swallowed or absorbed through skin. Avoid any skin contact. Effects of contact or inhalation may be delayed. Fire may produce irritating, corrosive and/or toxic gases. Runoff from fire control or dilution water may be corrosive and/or toxic and cause pollution.

Move victim to fresh air. Call 911 or emergency medical service. Give artificial respiration if victim is not breathing. Do not use mouth-to-mouth method if victim ingested or inhaled the substance;give artificial respiration with the aid of a pocket mask equipped with a one-way valve or other proper respiratory medical device. Administer oxygen if breathing is difficult. Remove and isolate contaminated clothing and shoes. In case of contact with substance, immediately flush skin or eyes with running water for at least 20 minutes. For minor skin contact, avoid spreading material on unaffected skin. Keep victim warm and quiet. Effects of exposure (inhalation, ingestion or skin contact) to substance may be delayed. Ensure that medical personnel are aware of the material(s) involved and take precautions to protect themselves.

The maximum tolerated human exposure to this agent has not been delineated. ANIMAL STUDIES - A daily dose of pindone (administered for 5 days) of 0.3 mg/kg for dogs and 2.5 mg/kg for chickens resulted in significant elevations in prothrombin time (more than double baseline values), but no clinical signs of anticoagulant poisoning (Martin et al, 1991).
