NICKELOUS SULFATE is used as a mordant in dyeing and printing fabrics, in blackening zinc and brass, in plating baths, and as a chemical intermediate for other nickel compounds (HSDB, 1996; (Budavari, 1996).
NICKELOUS SULFATE (CAS 7786-81-4), also called NICKEL(II) SULFATE, is the anhydrous form of the commercial product, NICKELOUS SULFATE HEXAHYDRATE (CAS 10101-97-0). The latter compound loses its water at approximately 100 degrees C (Budavari, 1996). This review discusses all forms of nickelous sulfate. Anhydrous nickel sulfate is a yellow cuboidal solid which is soluble in water (29.3 g/100 mL at 0 degrees C), and is insoluble in alcohol, ether, or acetone (HSDB, 1996; (Lewis, 1993). The hexahydrate occurs in two crystalline solid forms: alpha- (blue to blue-green tetragonal crystals), and beta- (green transparent crystals), which become blue and opaque at room temperature (HSDB, 1996; (Budavari, 1996).

ACUTE CLINICAL EFFECTS
- Nickel salts may act as emetics, producing nausea, vomiting, and diarrhea. Although oral toxicity is relatively low, systemic effects including capillary damage (especially in the brain and adrenals), kidney damage, myocardial depression, and central nervous system depression have been listed as effects of acute exposure to nickel salts (HSDB, 1996).
- Nickel sulfate is rapidly absorbed through intact skin in humans and experimental animals (Friberg et al, 1986).
- In rats, acute exposure to nickel sulfate produced excitation and increased motor activity, followed by central nervous system depression (Itskova, 1969).
CHRONIC CLINICAL EFFECTS
- ALLERGIC CONTACT DERMATITIS ("nickel itch"), is the most frequent effect of repeated skin exposure to nickel salts (HSDB, 1996; (Clayton & Clayton, 1994). The prevalence of nickel sensitivity in the general population is 2.5 to 5 percent (ACGIH, 1992). Nickel sulfate is used as the standard allergen for nickel sensitivity in patch tests and inhalation challenge testing.
- In North America, nickel sulfate is one of the most frequently detected sensitizers (Rudner, 1973). Nickel sensitivity may be permanent (Hathaway et al, 1991). The median duration of positive patch test reactions to nickelous sulfate is 9 days. There were no significant differences in dermal concentrations of nickel or in immuno-cytochemical parameters between subjects with long-lasting reactions and those who resolved normally (Handley et al, 1996).
- Nickel sulfate may also induce occupational asthma (Malo, 1982; Malo, 1985; Novey, 1983). Signs of respiratory allergy including nasal discharge, sneezing, and dryness and obstruction of the throat may be evident with chronic nickel sulfate exposure (pp 56-58).
- Chronic nickel sulfate exposure may produce eye irritation and excess tearing (HSDB, 1996).
- In chronically exposed experimental animals, nickel sulfate in the drinking water produced severe myocardial and liver damage in rabbits after 60 days (HSDB, 1996). Liver damage was also produced in rats exposed to nickel sulfate by skin painting (Mathur, 1977). Inhalation of nickel sulfate aerosol reduced the resistance of mice to pulmonary bacterial infection (Adkins, 1979).
- Highest concentrations of nickel were found in liver > kidney = whole blood = serum > testes > urine, in male rats exposed to nickel sulfate in the drinking water for 6 months. Concentrations were similar to those found after 3 months of exposure, and probably reflected steady-state levels (Severa et al, 1995). Low grade renal damage was reported when 100 mg/L of nickel sulfate in drinking water was provided to rats (Vyskocil et al, 1994).
