It is produced by reduction of 1,3,5-trimethyl-2-nitrobenzene, which is obtained by nitration of mesitylene under special conditions (HSDB, 2003).
-CLINICAL EFFECTS
GENERAL CLINICAL EFFECTS
- Aminomesitylene exposure causes eye and skin irritation. Chronic workplace exposure has been associated with various vision changes. Dermal sensitization may occur. Methemoglobinemia has not been reported in exposed humans, but exposure to other aniline compounds has caused this effect.

TOXIC; inhalation, ingestion or skin contact with material may cause severe injury or death. Contact with molten substance may cause severe burns to skin and eyes. Avoid any skin contact. Effects of contact or inhalation may be delayed. Fire may produce irritating, corrosive and/or toxic gases. Runoff from fire control or dilution water may be corrosive and/or toxic and cause pollution.
ACUTE CLINICAL EFFECTS
- Exposure to aminomesitylene may cause eye, skin, and respiratory or gastrointestinal tract irritation (RTECS, 1997; (Grant, 1986; Lewis, 1992).
- Methemoglobinemia has not been reported in exposed humans, but as exposure to many aniline compounds has caused this effect, it should be suspected in patients with significant exposure. Methemoglobinemia produces a central "chocolate-brown" cyanosis that does not improve with inhalation of 100 percent supplemental oxygen and signs and symptoms of hypoxia (Hall et al, 1986).
- Somnolence and gastrointestinal tract ulceration or bleeding have been reported in exposed experimental animals (RTECS, 1997).
CHRONIC CLINICAL EFFECTS
- Workers exposed to both aminomesitylene and aniline for 11 to 20 years were reported to develop vision effects including narrowed peripheral vision fields, increased size of the blind spot, and decreased photosensitivity (Grant, 1986). Changes in the morphology of the retina have been seen with aminomesitylene exposure in experimental animals (Grant, 1986).
- Cirrhosis of the liver occurred in male rats exposed to aminomesitylene in chronic dietary studies (Weisburger et al, 1978). Skin sensitization may also occur (EPA, 1985).
-FIRST AID
FIRST AID AND PREHOSPITAL TREATMENT
Exposed individuals should have a careful, thorough medical history and physical examination performed, looking for any abnormalities. Exposure to chemicals with a strong odor often results in such nonspecific symptoms as headache, dizziness, weakness, and nausea.
Emesis should not be induced. DILUTION: If no respiratory compromise is present, administer milk or water as soon as possible after ingestion. Dilution may only be helpful if performed in the first seconds to minutes after ingestion. The ideal amount is unknown; no more than 8 ounces (240 mL) in adults and 4 ounces (120 mL) in children is recommended to minimize the risk of vomiting. GASTRIC LAVAGE: Consider after ingestion of a potentially life-threatening amount of poison if it can be performed soon after ingestion (generally within 1 hour). Protect airway by placement in the head down left lateral decubitus position or by endotracheal intubation. Control any seizures first. Observe patients with ingestion carefully for the possible development of esophageal or gastrointestinal tract irritation or burns. If signs or symptoms of esophageal irritation or burns are present, consider endoscopy to determine the extent of injury.
INHALATION: Move patient to fresh air. Monitor for respiratory distress. If cough or difficulty breathing develops, evaluate for respiratory tract irritation, bronchitis, or pneumonitis. Administer oxygen and assist ventilation as required. Treat bronchospasm with an inhaled beta2-adrenergic agonist. Consider systemic corticosteroids in patients with significant bronchospasm. If bronchospasm and wheezing occur, consider treatment with inhaled sympathomimetic agents. ACUTE LUNG INJURY: Maintain ventilation and oxygenation and evaluate with frequent arterial blood gases and/or pulse oximetry monitoring. Early use of PEEP and mechanical ventilation may be needed.
DECONTAMINATION: Remove contaminated clothing and jewelry and place them in plastic bags. Wash exposed areas with soap and water for 10 to 15 minutes with gentle sponging to avoid skin breakdown. A physician may need to examine the area if irritation or pain persists (Burgess et al, 1999). Some chemicals can produce systemic poisoning by absorption through intact skin. Carefully observe patients with dermal exposure for the development of any systemic signs or symptoms and administer symptomatic treatment as necessary. Treat dermal irritation or burns with standard topical therapy. Patients developing dermal hypersensitivity reactions may require treatment with systemic or topical corticosteroids or antihistamines.
DECONTAMINATION: Remove contact lenses and irrigate exposed eyes with copious amounts of room temperature 0.9% saline or water for at least 15 minutes. If irritation, pain, swelling, lacrimation, or photophobia persist after 15 minutes of irrigation, the patient should be seen in a healthcare facility. Patients with any visual or ocular complaints following chronic exposure should have a thorough ophthalmologic evaluation.

Move victim to fresh air. Call 911 or emergency medical service. Give artificial respiration if victim is not breathing. Do not use mouth-to-mouth method if victim ingested or inhaled the substance; give artificial respiration with the aid of a pocket mask equipped with a one-way valve or other proper respiratory medical device. Administer oxygen if breathing is difficult. Remove and isolate contaminated clothing and shoes. In case of contact with substance, immediately flush skin or eyes with running water for at least 20 minutes. For minor skin contact, avoid spreading material on unaffected skin. Keep victim warm and quiet. Effects of exposure (inhalation, ingestion or skin contact) to substance may be delayed. Ensure that medical personnel are aware of the material(s) involved and take precautions to protect themselves.

Move patients with inhalation exposure from the toxic environment and administer 100% supplemental oxygen with assisted ventilation as required. Exposed skin and eyes should be thoroughly flushed. As irritation or burns of the esophagus could occur with ingestion, emesis should not be induced. Immediate dilution with milk or water may be beneficial. If cyanosis is present, obtain and monitor methemoglobin levels. Treatment with 100% oxygen and intravenous methylene blue could be required. METHEMOGLOBINEMIA: Determine the methemoglobin concentration and evaluate the patient for clinical effects of methemoglobinemia (ie, dyspnea, headache, fatigue, CNS depression, tachycardia, metabolic acidosis). Treat patients with symptomatic methemoglobinemia with methylene blue (this usually occurs at methemoglobin concentrations above 20% to 30%, but may occur at lower methemoglobin concentrations in patients with anemia, or underlying pulmonary or cardiovascular disorders). Administer oxygen while preparing for methylene blue therapy. METHYLENE BLUE: INITIAL DOSE/ADULT OR CHILD: 1 mg/kg IV over 5 to 30 minutes; a repeat dose of up to 1 mg/kg may be given 1 hour after the first dose if methemoglobin levels remain greater than 30% or if signs and symptoms persist. NOTE: Methylene blue is available as follows: 50 mg/10 mL (5 mg/mL or 0.5% solution) single-dose ampules and 10 mg/1 mL (1% solution) vials. Additional doses may sometimes be required. Improvement is usually noted shortly after administration if diagnosis is correct. Consider other diagnoses or treatment options if no improvement has been observed after several doses. If intravenous access cannot be established, methylene blue may also be given by intraosseous infusion. Methylene blue should not be given by subcutaneous or intrathecal injection. NEONATES: DOSE: 0.3 to 1 mg/kg. Concomitant use of methylene blue with serotonergic drugs, including serotonin reuptake inhibitors (SRIs), selective serotonin reuptake inhibitors (SSRIs), serotonin and norepinephrine reuptake inhibitors (SNRIs), tricyclic antidepressants (TCAs), norepinephrine-dopamine reuptake inhibitors (NDRIs), triptans, and ergot alkaloids may increase the risk of potentially fatal serotonin syndrome.
Patients with hypersensitivity dermatitis may require treatment with corticosteroids or antihistamines. Removal from further exposure may be necessary.

Rabbit skin was moderately irritated when 500 milligrams of aminomesitylene were applied for 24 hours (RTECS , 1996). Rabbit eyes were severely irritated when 20 milligrams of aminomesitylene were applied for 24 hours (RTECS , 1996).

Aminomesitylene released to the air is expected to exist solely as a vapor, based on its estimated vapor pressure of 0.067 mmHg (at 25 degrees C). Degradation of the vapor through reaction with photochemically-produced hydroxyl radicals will occur. The rate constant for this reaction is estimated as 5.7 x 10(-11) cm(3)/molecule-sec (at 25 degrees C), which corresponds to an atmospheric half-life of 6.7 hours (at an atmospheric concentration of 5 x 10(5) hydroxyl radicals/cm(3)) (HSDB, 2004). Direct photolysis is not expected to occur, due to the lack of absorption at wavelengths >290 nm (HSDB, 2004).
SURFACE WATER In water, aminomesitylene is expected to adsorb slightly to sediment and suspended solids, based on its estimated organic carbon/water partition coefficient (Koc) of 198. Volatilization from water surfaces is expected to be significant, based on an estimated Henry's Law constant of 2.7 x 10(-6) atm-cm(3)/mol, but will be limited by adsorption of aminomesitylene to humate in the water column(HSDB, 2004). Volatilization half-lives from a model river (1 m deep, flowing at 1 m/sec, wind velocity of 3 m/sec) and model lake (1 m deep, flowing at 0.05 m/sec, wind velocity of 0.5 m/sec) have been estimated at 11 and 120 days, respectively (HSDB, 2004).
