m-Toluidine is used in the manufacture of certain dyes and other organic chemicals. Its use is limited to 12 dyes because of its nonplanar chemical configuration (HSDB , 1996; Clayton & Clayton, 1994).
-CLINICAL EFFECTS
GENERAL CLINICAL EFFECTS
- m-Toluidine is toxic by inhalation and ingestion, and can be absorbed through intact skin. It is a skin and eye irritant.
- CNS depression may occur following inhalation, ingestion, or cutaneous absorption. Cyanosis, methemoglobinemia, hematuria, and anuria may also be noted. Gastrointestinal symptoms include anorexia, nausea, vomiting, and diarrhea.
- Other symptoms may include defatting dermatitis, weakness, headache, drowsiness, dyspnea, hypotension, and jaundice.

SUMMARY
- FIRST AID - EMERGENCY RESPONSE GUIDEBOOK, GUIDE 153 (ERG, 2004)
Move victim to fresh air. Call 911 or emergency medical service. Give artificial respiration if victim is not breathing. Do not use mouth-to-mouth method if victim ingested or inhaled the substance; give artificial respiration with the aid of a pocket mask equipped with a one-way valve or other proper respiratory medical device. Administer oxygen if breathing is difficult. Remove and isolate contaminated clothing and shoes. In case of contact with substance, immediately flush skin or eyes with running water for at least 20 minutes. For minor skin contact, avoid spreading material on unaffected skin. Keep victim warm and quiet. Effects of exposure (inhalation, ingestion or skin contact) to substance may be delayed. Ensure that medical personnel are aware of the material(s) involved and take precautions to protect themselves.
FIRST AID EYE EXPOSURE - Immediately wash the eyes with large amounts of water, occasionally lifting the lower and upper lids. Get medical attention immediately. Contact lenses should not be worn when working with this chemical. DERMAL EXPOSURE - Immediately wash the contaminated skin with soap and water. If this chemical penetrates the clothing, immediately remove the clothing, wash the skin with soap and water, and get medical attention promptly. INHALATION EXPOSURE - Move the exposed person to fresh air at once. If breathing has stopped, perform artificial respiration. Keep the affected person warm and at rest. Get medical attention as soon as possible. ORAL EXPOSURE - If this chemical has been swallowed, get medical attention immediately. TARGET ORGANS - Eyes, skin, blood, and cardiovascular system (National Institute for Occupational Safety and Health, 2007).
GENERAL Move victims of inhalation exposure from the toxic environment and administer 100% humidified supplemental oxygen with assisted ventilation as required. Exposed skin and eyes should be copiously flushed with water. Because of the potential for rapid onset of CNS depression or seizures with possible aspiration of gastric contents, EMESIS SHOULD NOT BE INDUCED. Cautious gastric lavage followed by administration of activated charcoal may be of benefit if the patient is seen soon after the exposure.
INHALATION EXPOSURE INHALATION: Move patient to fresh air. Monitor for respiratory distress. If cough or difficulty breathing develops, evaluate for respiratory tract irritation, bronchitis, or pneumonitis. Administer oxygen and assist ventilation as required. Treat bronchospasm with an inhaled beta2-adrenergic agonist. Consider systemic corticosteroids in patients with significant bronchospasm. If bronchospasm and wheezing occur, consider treatment with inhaled sympathomimetic agents. Carefully observe patients with inhalation exposure for the development of any systemic signs or symptoms and administer symptomatic treatment as necessary.
DERMAL EXPOSURE DECONTAMINATION: Remove contaminated clothing and jewelry and place them in plastic bags. Wash exposed areas with soap and water for 10 to 15 minutes with gentle sponging to avoid skin breakdown. A physician may need to examine the area if irritation or pain persists (Burgess et al, 1999). Treat dermal irritation or burns with standard topical therapy. Patients developing dermal hypersensitivity reactions may require treatment with systemic or topical corticosteroids or antihistamines.
EYE EXPOSURE DECONTAMINATION: Remove contact lenses and irrigate exposed eyes with copious amounts of room temperature 0.9% saline or water for at least 15 minutes. If irritation, pain, swelling, lacrimation, or photophobia persist after 15 minutes of irrigation, the patient should be seen in a healthcare facility.
ORAL EXPOSURE Because of the potential for gastrointestinal tract irritation or CNS depression, do not induce emesis. Significant esophageal or gastrointestinal tract irritation or burns may occur following ingestion. The possible benefit of early removal of some ingested material by cautious gastric lavage must be weighed against potential complications of bleeding or perforation. GASTRIC LAVAGE: Consider after ingestion of a potentially life-threatening amount of poison if it can be performed soon after ingestion (generally within 1 hour). Protect airway by placement in the head down left lateral decubitus position or by endotracheal intubation. Control any seizures first. ACTIVATED CHARCOAL: Administer charcoal as a slurry (240 mL water/30 g charcoal). Usual dose: 25 to 100 g in adults/adolescents, 25 to 50 g in children (1 to 12 years), and 1 g/kg in infants less than 1 year old. METHEMOGLOBINEMIA: Determine the methemoglobin concentration and evaluate the patient for clinical effects of methemoglobinemia (ie, dyspnea, headache, fatigue, CNS depression, tachycardia, metabolic acidosis). Treat patients with symptomatic methemoglobinemia with methylene blue (this usually occurs at methemoglobin concentrations above 20% to 30%, but may occur at lower methemoglobin concentrations in patients with anemia, or underlying pulmonary or cardiovascular disorders). Administer oxygen while preparing for methylene blue therapy. METHYLENE BLUE: INITIAL DOSE/ADULT OR CHILD: 1 mg/kg IV over 5 to 30 minutes; a repeat dose of up to 1 mg/kg may be given 1 hour after the first dose if methemoglobin levels remain greater than 30% or if signs and symptoms persist. NOTE: Methylene blue is available as follows: 50 mg/10 mL (5 mg/mL or 0.5% solution) single-dose ampules and 10 mg/1 mL (1% solution) vials. Additional doses may sometimes be required. Improvement is usually noted shortly after administration if diagnosis is correct. Consider other diagnoses or treatment options if no improvement has been observed after several doses. If intravenous access cannot be established, methylene blue may also be given by intraosseous infusion. Methylene blue should not be given by subcutaneous or intrathecal injection. NEONATES: DOSE: 0.3 to 1 mg/kg. Concomitant use of methylene blue with serotonergic drugs, including serotonin reuptake inhibitors (SRIs), selective serotonin reuptake inhibitors (SSRIs), serotonin and norepinephrine reuptake inhibitors (SNRIs), tricyclic antidepressants (TCAs), norepinephrine-dopamine reuptake inhibitors (NDRIs), triptans, and ergot alkaloids may increase the risk of potentially fatal serotonin syndrome. Observe patients with ingestion carefully for the possible development of esophageal or gastrointestinal tract irritation or burns. If signs or symptoms of esophageal irritation or burns are present, consider endoscopy to determine the extent of injury.

m-Toluidine has not been as extensively tested as o-toluidine, but similar effects of exposure are expected. Both isomers have similar toxicity profiles and dose ranges (Hathaway, 1991). No epidemiological studies have been published on workers exposed exclusively to m-toluidine (Clayton & Clayton, 1994). Exposure to 10 ppm of mixed isomers of toluidine for more than a short time caused symptoms of illness, while 40 ppm for 1 hour reportedly caused severe toxic effects (ACGIH, 1991). m-Toluidine has not been shown to be carcinogenic in any animal studies (Hathaway, 1991).

m-Toluidine should be stored in a dry, cool, well-ventilated location, away from potential sources of fire. Outside detached storage away from other storage areas is preferable (ITI, 1988).

Wear positive pressure self-contained breathing apparatus (SCBA). Wear chemical protective clothing that is specifically recommended by the manufacturer. It may provide little or no thermal protection. Structural firefighters' protective clothing provides limited protection. fire situations ONLY; it is not effective in spill situations where direct contact with the substance is possible.
- Personnel should stay upwind and avoid breathing m-toluidine vapors. Positive pressure self-contained breathing apparatus and appropriate chemical protective clothing, including butyl rubber boots, gloves, and safety glasses, should be worn when contact with m-toluidine is expected (AAR, 1994; (ITI, 1988).

POTENTIAL FIRE OR EXPLOSION HAZARDS - EMERGENCY RESPONSE GUIDEBOOK, GUIDE 153 (ERG, 2004) Combustible material: may burn but does not ignite readily. When heated, vapors may form explosive mixtures with air: indoors, outdoors and sewers explosion hazards. Those substances designated with a "P" may polymerize explosively when heated or involved in a fire. Contact with metals may evolve flammable hydrogen gas. Containers may explode when heated. Runoff may pollute waterways. Substance may be transported in a molten form.
m-Toluidine is combustible when exposed to heat or flame (Lewis, 1996). Water applied from as far away as possible should be used in flooding quantities as fog and to cool containers (AAR, 1994).

REACTIVITY HAZARD
- m-Toluidine may react vigorously with oxidizing materials (Lewis, 1996).
- Potassium iodide solution added to diazotized m-toluidine may cause the container to erupt (Bretherick, 1995).
- Very toxic fumes of nitrogen oxides are released when m-toluidine is heated to decomposition (Lewis, 1996).
- m-Toluidine is incompatible with acids (NIOSH , 1996).
- Aromatic amines such as toluidines in triethylamine solution are rapidly ignited by red fuming nitric acid at minus 76 degrees or lower (NFPA, 1994).

RECOMMENDED PROTECTIVE CLOTHING - EMERGENCY RESPONSE GUIDEBOOK, GUIDE 153 (ERG, 2004) Wear positive pressure self-contained breathing apparatus (SCBA). Wear chemical protective clothing that is specifically recommended by the manufacturer. It may provide little or no thermal protection. Structural firefighters' protective clothing provides limited protection. fire situations ONLY; it is not effective in spill situations where direct contact with the substance is possible.
Sparks, flames, and other ignition sources should be kept away from spills involving m-toluidine (AAR, 1994). Spills involving m-toluidine may be covered with a 9:1 mixture of sand and soda ash. The material may then be incinerated carefully after mixing with combustible material such as paper (ITI, 1988).
Biological treatment may be a possible method for removing m-toluidine from wastewater streams (HSDB , 1996). Waste management activities associated with material disposition are unique to individual situations. Proper waste characterization and decisions regarding waste management should be coordinated with the appropriate local, state, or federal authorities to ensure compliance with all applicable rules and regulations.
Controlled incineration may be used to dispose of waste streams containing m-toluidine. The incinerator should include effluent gas scrubbers and/or thermal devices to remove nitrogen oxides from waste streams (HSDB , 1996). m-Toluidine may be dissolved in a combustible solvent such as alcohol or benzene and then sprayed into an incinerator equipped with an afterburner and scrubber (ITI, 1988).

- m-Toluidine may be released to wastewater streams during its manufacture and during its use in the production of dyes and other chemicals (HSDB , 1996).
ENVIRONMENTAL FATE AND KINETICS
SURFACE WATER m-Toluidine released to water is expected to rapidly biodegrade. It may also be photooxidized or may be oxidized by dissolved oxygen, ferric and other metal ions, and humic materials found in natural surface waters. The photooxidation half-life in distilled water is approximately 68 hours (HSDB , 1996). The volatilization half-life of m-toluidine from a model river is estimated to be 16.3 days. The rate is controlled by the diffusion through air (HSDB , 1996).
ABIOTIC DEGRADATION
- Direct photolysis by sunlight is expected to be an important fate process for m-toluidine, since it has a UV absorption band at 286 nm. Colored degradation products are formed when light and oxygen are present. The photochemical half-life was approximately 68 hours in one study using a fluorochemical lamp (HSDB , 1996).
- Soil- and clay-catalyzed free radical oxidation is also a possible fate process for m-toluidine (HSDB , 1996).
BIODEGRADATION
- m-Toluidine has been shown to be readily biodegradable in various screening tests and microbial degradation is expected to be an important fate process for m-toluidine in natural waters and soils (HSDB , 1996).
