Fluoroacetyl chloride is an acid chloride compound. No information on the appearance or odor of fluoroacetyl chloride were found in available references at the time of this review. Few specific data were available about the toxicity of fluoroacetyl chloride; its toxicity is expected to be similar to that of FLUOROACETATE. This review is based on the properties of fluoroacetate (as SODIUM FLUOROACETATE). Effects attributed specifically to fluoroacetyl chloride are identified.
-CLINICAL EFFECTS
GENERAL CLINICAL EFFECTS
- Fluoroacetyl chloride is the acid chloride of sodium fluoroacetate. Fluoro ester compounds can be hydrolyzed directly to fluoroacetate. Little specific data were available specifically regarding the toxicity of fluoroacetyl chloride; its toxicity is expected to be similar to that of FLUOROACETATE.
The following review discusses the toxicity and treatment of poisoning with FLUOROACETATE. No data on the appearance, odor, or uses of fluoroacetyl chloride were found in available references at the time of this review, FLUOROACETYL CHLORIDE is irritating or corrosive to the skin and eyes, and could be irritating to the GI tract if ingested. No data are available regarding whether this compound can be absorbed through intact skin; it may be absorbed following ingestion and inhalation.
- Clinical effects are usually seen within 1/2 hour of exposure. Symptoms of nausea, vomiting, excessive salivation, abdominal pain, numbness, a tingling sensation, and apprehension are seen initially, and may last for up to 6 hours. Muscular twitching, blurred vision, and hypotension may develop.
Severe effects such as coma, convulsions, and cardiac arrhythmias may be delayed in onset as long as 20 hours. The cardiac effects noted include tachycardia, ventricular fibrillation, and sudden onset of asystole. Death may occur from respiratory depression and hypoxia during convulsions or cardiac arrest. Neurologic sequelae and acute renal failure have been described after acute poisoning. Metabolic acidosis, hyperglycemia, hyperuricemia, elevated levels of hepatic transaminases, and elevated serum creatinine may occur in fluoroacetate poisoning.
- At least one case of severe poisoning with numbness and tingling of the face, excessive salivation, blurred vision, peripheral paresthesias, convulsions, and coma has occurred from inhalation and dermal contact with fluoroacetate. In general, fluoroacetate is absorbed following ingestion and inhalation, but not through intact skin.
- Fluoroacetate mimics acetic acid and reacts with coenzyme A and oxaloacetic acid, forming fluorocitric acid which enters and blocks the Kreb's cycle, allowing accumulation of citric acid.
- Fluoroacetyl chloride release toxic and irritating fumes of chlorides and fluorides when heated to decomposition. Inhalation exposure to such products of combustion would be predicted to result in respiratory tract irritation with bronchospasm, chemical pneumonitis, or noncardiogenic pulmonary edema.

ACUTE CLINICAL EFFECTS
- Based on its acute inhalation LC50 of 200 mg/m(3)/10 minutes in mice (RTECS , 1993), fluoroacetyl chloride is a moderately toxic substance. It may be absorbed following ingestion and inhalation (Sax & Lewis, 1989; EPA, 1985). No data are available about whether this compound can be absorbed through intact skin.
- Fluoroacetyl chloride is irritating or corrosive to the skin and eyes (EPA, 1985), and could be irritating to the GI tract if ingested.
- Fluoroacetate is very rapidly absorbed. A milligram of pure fluoroacetate is probably enough to cause severe toxicity, and less may be toxic. A dose of 2 to 10 mg/kg (140 to 700 mg for a 70-kg adult) may be fatal (Egekeze & Oehme, 1979; Baselt & Cravey, 1989). One death due to subacute fluoroacetate poisoning has been reported (Peters et al, 1981).

- Severe effects of fluoroacetate poisoning such as coma, convulsions, and cardiac arrhythmias may be delayed in onset as long as 20 hours (Reigart et al, 1975; HSDB , 1993). Death may occur from respiratory depression and hypoxia during convulsions or cardiac arrest (HSDB , 1993).
- Acute fluoroacetate poisoning can affect the heart. Cardiac effects noted include tachycardia, ventricular fibrillation, and sudden onset of asystole (HSDB , 1993; Ellenhorn & Barceloux, 1988). Other effects described with acute fluoroacetate poisoning are delayed neurotoxicity, acute kidney failure, metabolic acidosis, hyperglycemia, hyperuricemia, elevated levels of hepatic transaminases, and elevated serum creatinine (McTaggart, 1970; Trabes et al, 1983; Baselt & Cravey, 1989; Ellenhorn & Barceloux, 1988; Chung, 1984).
- Fluoroacetate mimics acetic acid and reacts with coenzyme A and oxaloacetic acid, forming fluorocitric acid which enters and blocks the Krebs cycle, with accumulation of CITRIC ACID as the result (HSDB , 1993; Baselt & Cravey, 1989).
CHRONIC CLINICAL EFFECTS
- At the time of this review, no studies on chronic exposure to fluoroacetyl chloride in humans or experimental animals were found.
- Neurologic, liver, kidney, and thyroid dysfunction were seen in an exterminator who was repeatedly exposed to fluoroacetate during a 10-year period and in a rabbiter who was repeatedly exposed to sodium fluoroacetate (Parkin et al, 1977; Proctor et al, 1988). No other reports of chronic exposure to fluoroacetate were found.

MINIMUM LETHAL EXPOSURE
A milligram of pure compound is probably enough to cause severe toxicity, and less may be toxic. Extrapolation of experimental animal toxicity data to humans indicates that a dose of 2 to 10 mg/kg (140 to 700 mg for a 70-kg adult) may be fatal (Egekeze & Oehme, 1979; Baselt & Cravey, 1989).
A death was reported after ingestion of 465 mg or more in an adult (Harrisson et al, 1952). Prior to 1955, there were 23 cases of poisoning in humans, 17 with fatal outcomes (Brockmann et al, 1955).
