USES/FORMS/SOURCES
Diphenylamine is available in the technical, refined, flake or fused grades (Lewis, 1993). 4-Biphenylamine, a potent carcinogen, may be present as an impurity in some preparations (HSDB , 1998).
Diphenylamine may be produced through the combination of aniline and aniline hydrochloride with heat. This compound is used to manufacture dyes, pharmaceuticals, and pesticides. It is used as a stabilizer for nitrocellulose explosives, celluloid, and plastics, including solid rocket propellants. Diphenylamine is used as an antioxidant and to preserve stored apples. It may also be used as a fungicide and antihelmintic and in tests for nitrate or nitrite poisoning. In analytical chemistry it is used to detect nitrates, chlorates and other oxidizing substances (ACGIH, 1991; Budavari, 1996; Lewis, 1993; OHM/TADS , 1998).
-CLINICAL EFFECTS
GENERAL CLINICAL EFFECTS
- Diphenylamine is readily absorbed orally and through the respiratory tract and skin. It is irritating to the eyes, skin, and mucous membranes. Signs and symptoms may include hypertension, tachycardia, cough, sneezing, urinary symptoms, anorexia, eczema, and methemoglobinemia.

Move victim to fresh air. Call 911 or emergency medical service. Give artificial respiration if victim is not breathing. Do not use mouth-to-mouth method if victim ingested or inhaled the substance; give artificial respiration with the aid of a pocket mask equipped with a one-way valve or other proper respiratory medical device. Administer oxygen if breathing is difficult. Remove and isolate contaminated clothing and shoes. In case of contact with substance, immediately flush skin or eyes with running water for at least 20 minutes. For minor skin contact, avoid spreading material on unaffected skin. Keep victim warm and quiet. Effects of exposure (inhalation, ingestion or skin contact) to substance may be delayed. Ensure that medical personnel are aware of the material(s) involved and take precautions to protect themselves.
FIRST AID EYE EXPOSURE - Immediately wash the eyes with large amounts of water, occasionally lifting the lower and upper lids. Get medical attention immediately. Contact lenses should not be worn when working with this chemical. DERMAL EXPOSURE - Promptly wash the contaminated skin with soap and water. If this chemical penetrates the clothing, promptly remove the clothing and wash the skin with soap and water. Get medical attention promptly. INHALATION EXPOSURE - Move the exposed person to fresh air at once. If breathing has stopped, perform artificial respiration. Keep the affected person warm and at rest. Get medical attention as soon as possible. ORAL EXPOSURE - If this chemical has been swallowed, get medical attention immediately. TARGET ORGANS - Eyes, skin, respiratory system, cardiovascular system, blood, bladder, and reproductive system (National Institute for Occupational Safety and Health, 2007).
GENERAL Move victims of inhalation exposure from the toxic environment and administer 100% humidified supplemental oxygen with assisted ventilation as required. Exposed skin and eyes should be copiously flushed with water. Ingestion may result in significant esophageal or gastrointestinal tract irritation or burns, and EMESIS SHOULD NOT BE INDUCED. Cautious gastric lavage followed by administration of activated charcoal may be of benefit if the patient is seen soon after the exposure.
INHALATION EXPOSURE INHALATION: Move patient to fresh air. Monitor for respiratory distress. If cough or difficulty breathing develops, evaluate for respiratory tract irritation, bronchitis, or pneumonitis. Administer oxygen and assist ventilation as required. Treat bronchospasm with an inhaled beta2-adrenergic agonist. Consider systemic corticosteroids in patients with significant bronchospasm. If bronchospasm and wheezing occur, consider treatment with inhaled sympathomimetic agents. Carefully observe patients with inhalation exposure for the development of any systemic signs or symptoms and administer symptomatic treatment as necessary. METHEMOGLOBINEMIA: Determine the methemoglobin concentration and evaluate the patient for clinical effects of methemoglobinemia (ie, dyspnea, headache, fatigue, CNS depression, tachycardia, metabolic acidosis). Treat patients with symptomatic methemoglobinemia with methylene blue (this usually occurs at methemoglobin concentrations above 20% to 30%, but may occur at lower methemoglobin concentrations in patients with anemia, or underlying pulmonary or cardiovascular disorders). Administer oxygen while preparing for methylene blue therapy. METHYLENE BLUE: INITIAL DOSE/ADULT OR CHILD: 1 mg/kg IV over 5 to 30 minutes; a repeat dose of up to 1 mg/kg may be given 1 hour after the first dose if methemoglobin levels remain greater than 30% or if signs and symptoms persist. NOTE: Methylene blue is available as follows: 50 mg/10 mL (5 mg/mL or 0.5% solution) single-dose ampules and 10 mg/1 mL (1% solution) vials. Additional doses may sometimes be required. Improvement is usually noted shortly after administration if diagnosis is correct. Consider other diagnoses or treatment options if no improvement has been observed after several doses. If intravenous access cannot be established, methylene blue may also be given by intraosseous infusion. Methylene blue should not be given by subcutaneous or intrathecal injection. NEONATES: DOSE: 0.3 to 1 mg/kg. Concomitant use of methylene blue with serotonergic drugs, including serotonin reuptake inhibitors (SRIs), selective serotonin reuptake inhibitors (SSRIs), serotonin and norepinephrine reuptake inhibitors (SNRIs), tricyclic antidepressants (TCAs), norepinephrine-dopamine reuptake inhibitors (NDRIs), triptans, and ergot alkaloids may increase the risk of potentially fatal serotonin syndrome.
DERMAL EXPOSURE DECONTAMINATION: Remove contaminated clothing and jewelry and place them in plastic bags. Wash exposed areas with soap and water for 10 to 15 minutes with gentle sponging to avoid skin breakdown. A physician may need to examine the area if irritation or pain persists (Burgess et al, 1999). Treat dermal irritation or burns with standard topical therapy. Patients developing dermal hypersensitivity reactions may require treatment with systemic or topical corticosteroids or antihistamines. Some chemicals can produce systemic poisoning by absorption through intact skin. Carefully observe patients with dermal exposure for the development of any systemic signs or symptoms and administer symptomatic treatment as necessary. METHEMOGLOBINEMIA: Determine the methemoglobin concentration and evaluate the patient for clinical effects of methemoglobinemia (ie, dyspnea, headache, fatigue, CNS depression, tachycardia, metabolic acidosis). EYE EXPOSURE DECONTAMINATION: Remove contact lenses and irrigate exposed eyes with copious amounts of room temperature 0.9% saline or water for at least 15 minutes. If irritation, pain, swelling, lacrimation, or photophobia persist after 15 minutes of irrigation, the patient should be seen in a healthcare facility.
ORAL EXPOSURE Because of the potential for gastrointestinal tract irritation, DO NOT induce emesis. Significant esophageal or gastrointestinal tract irritation or burns may occur following ingestion. The possible benefit of early removal of some ingested material by cautious gastric lavage must be weighed against potential complications of bleeding or perforation. PREHOSPITAL ACTIVATED CHARCOAL ADMINISTRATION Consider prehospital administration of activated charcoal as an aqueous slurry in patients with a potentially toxic ingestion who are awake and able to protect their airway. Activated charcoal is most effective when administered within one hour of ingestion. Administration in the prehospital setting has the potential to significantly decrease the time from toxin ingestion to activated charcoal administration, although it has not been shown to affect outcome (Alaspaa et al, 2005; Thakore & Murphy, 2002; Spiller & Rogers, 2002). In patients who are at risk for the abrupt onset of seizures or mental status depression, activated charcoal should not be administered in the prehospital setting, due to the risk of aspiration in the event of spontaneous emesis. The addition of flavoring agents (cola drinks, chocolate milk, cherry syrup) to activated charcoal improves the palatability for children and may facilitate successful administration (Guenther Skokan et al, 2001; Dagnone et al, 2002).
ACTIVATED CHARCOAL: Administer charcoal as a slurry (240 mL water/30 g charcoal). Usual dose: 25 to 100 g in adults/adolescents, 25 to 50 g in children (1 to 12 years), and 1 g/kg in infants less than 1 year old. Observe patients with ingestion carefully for the possible development of esophageal or gastrointestinal tract irritation or burns. If signs or symptoms of esophageal irritation or burns are present, consider endoscopy to determine the extent of injury. Carefully observe patients with ingestion exposure for the development of any systemic signs or symptoms and administer symptomatic treatment as necessary. METHEMOGLOBINEMIA: Determine the methemoglobin concentration and evaluate the patient for clinical effects of methemoglobinemia (ie, dyspnea, headache, fatigue, CNS depression, tachycardia, metabolic acidosis). Diphenylamine should be stored in an area that is kept cool and well-ventilated. It should also be protected from light since it discolors on exposure. Storage areas should also be free from ignition sources or open flames. Diphenylamine may react with oxidizers and should be separated from them in storage (HSDB , 1998; Sittig, 1991). Personnel who may be working with diphenylamine or exposed to it in the workplace should be equipped with proper personal protective equipment such as rubber gloves, protective clothing, respiratory protection, and safety glasses (ITI, 1995; OHM/TADS , 1998).

If this compound is stored in steel equipment for a long period of time, it should be kept at temperatures below 65 degrees C (OHM/TADS , 1998; Sittig, 1991).

Wear positive pressure self-contained breathing apparatus (SCBA). Wear chemical protective clothing that is specifically recommended by the manufacturer. It may provide little or no thermal protection. Structural firefighters' protective clothing provides limited protection in fire situations ONLY; it is not effective in spill situations where direct contact with the substance is possible.
- Diphenylamine exposure can take place through inhalation, skin absorption, eye contact, or ingestion, so workers should be equipped with proper personal protective equipment to prevent exposure. POTENTIAL FIRE OR EXPLOSION HAZARDS - EMERGENCY RESPONSE GUIDEBOOK, GUIDE 134 (ERG, 2004) Flammable/combustible material. May be ignited by heat, sparks or flames. When heated, vapors may form explosive mixtures with air: indoors, outdoors and sewers explosion hazards. Contact with metals may evolve flammable hydrogen gas. Containers may explode when heated.
Diphenylamine is a compound that is combustible, but generally requires heating before it poses a fire hazard. Fires involving diphenylamine should be extinguished with dry chemicals, foam, or carbon dioxide. Foam extinguishing agents and water can cause frothing and may not be effective. Containers should be kept cool with water if exposed to fire (CHRIS , 1998; OHM/TADS , 1998; Sittig, 1991). If diphenylamine is involved in a fire, it will give off toxic oxides of nitrogen. Therefore, personnel fighting fires should be equipped with self-contained breathing apparatus and other protective equipment. Diphenylamine dust can be explosive if mixed with air and exposed to an ignition source (CHRIS , 1998).

SPILL OR LEAK PRECAUTIONS - EMERGENCY RESPONSE GUIDEBOOK, GUIDE 134 (ERG, 2004) Fully encapsulating, vapor protective clothing should be worn for spills and leaks with no fire. ELIMINATE all ignition sources (no smoking, flares, sparks or flames in immediate area). Stop leak if you can do it without risk. Do not touch damaged containers or spilled material unless wearing appropriate protective clothing. Prevent entry into waterways, sewers, basements or confined areas. Use clean non-sparking tools to collect material and place it into loosely covered plastic containers for later disposal.
RECOMMENDED PROTECTIVE CLOTHING - EMERGENCY RESPONSE GUIDEBOOK, GUIDE 134 (ERG, 2004) Wear positive pressure self-contained breathing apparatus (SCBA). Wear chemical protective clothing that is specifically recommended by the manufacturer. It may provide little or no thermal protection. Structural firefighters' protective clothing provides limited protection in fire situations ONLY; it is not effective in spill situations where direct contact with the substance is possible.
In case of a diphenylamine spill, persons not equipped with personal protective equipment should be prevented from entering the spill area. The area should be well ventilated and all ignition sources should be eliminated. Spilled material in powdered form should be cleaned up and put in sealed containers for appropriate disposal. Spilled diphenylamine should be isolated and contained by dams, or other impoundments to prevent its spread. In water, diphenylamine will sink and could pose a threat if it enters water intake systems (CHRIS , 1998; OHM/TADS , 1998) Sittig, 1998).
Waters contaminated with dilute diphenylamine may be treated using sorption on powdered activated carbon or filtration with granular activated carbon. Well-point collection and leachate treatment may be used to treat contaminated soils (OHM/TADS , 1998). Waste management activities associated with material disposition are unique to individual situations. Proper waste characterization and decisions regarding waste management should be coordinated with the appropriate local, state, or federal authorities to ensure compliance with all applicable rules and regulations.
Diphenylamine may be incinerated after mixing with a flammable solvent. The furnace should be equipped with an afterburner and scrubber. It may also be disposed of in an appropriate landfill (HSDB , 1998; Sittig, 1991).

SURFACE WATER Estimates for the half-life of diphenylamine in surface water range from 31 to 672 hours, while those for groundwater range from 336 to 1344 hours. In water, the aerobic half-life for biodegradation ranges from 168 to 672 hours. Estimates for aqueous anaerobic half-life range from 672 to 2688 hours. The photooxidation half-life in water ranges from 31 to 1740 hours (Howard et al, 1991).
