DDAC is used as a fungicide in antisapstain products in the forest industry (Henderson, 1992; Chen et al, 1995). DDAC is also an ingredient of antimicrobials and antiseptics, household cleaners, mildew preventatives, and biocidal water treatment (Henderson, 1992).
Didecyldimethylammonium chloride, also called DDAC or QUATERNIUM-12, is a quaternary ammonium compound with the properties of a cationic detergent. It is very soluble in water and is hydrolytically and photolytically stable (Henderson, 1992). The following review is based on properties of cationic detergents in general, with effects specific to DDAC noted. The toxicity of DDAC has been reviewed by Henderson (1992), using unpublished sources.

GENERAL CLINICAL EFFECTS

ACUTE CLINICAL EFFECTS
- At the time of this review, no information was found on effects of acute exposure to DDAC in humans. With an acute oral LD50 in rats of 84 mg/kg and 268 mg/kg mice, respectively, DDAC is a moderately to very toxic substance (RTECS , 1996).
- DDAC is a severe skin and eye irritant (Henderson, 1992). It is a severe skin irritant in rabbits in the standard Draize test (RTECS , 1996). The non-irritating concentration for rabbits was 0.005% DDAC (as 50% of a product called ALIQUAT 203) (Henderson, 1992).
- Human fatalities have occurred from ingestion of quaternary ammonium compounds, but the nature and severity of the response may vary with the compound and its concentration (Henderson, 1992). The degree of irritation or corrosion would increase with increasing concentration. Eye exposure to cationic detergents may result in mild discomfort to severe corneal damage. Mild to severe caustic burns of the lips, tongue, mouth, and throat have occurred with cationic detergent ingestion.
- Cationic detergents as a class can be dangerous or even fatal if ingested. Vomiting, diarrhea, hemorrhagic necrosis, peritonitis, dermal necrosis, dermatitis, pulmonary edema, hypotension, liver damage, metabolic acidosis, and CNS depression have been reported with ingestion of other cationic detergents (Adelson & Sunshine, 1952) van Berkel & de Wolff, 1988). Ingestion of cationic detergents may produce profound coma, seizures, and shock. Death may be due to respiratory paralysis.
- Acute exposure to DDAC by the oral route in rats and mice produced general depressed activity. Muscle weakness occurred in rats and mice exposed by the IP route. Convulsions or effect on seizure threshold and changes in the respiratory system were seen in guinea pigs exposed by the IP route (RTECS , 1996).
- (14C)-DDAC was not well absorbed from the gastrointestinal tract in rats. From 89 to 99% of the recovered radioactivity was in the feces, with less than 2.5% in the urine. None was exhaled as (14C)-CO2 within 24 hours after a single oral dose. DDAC is metabolized in rats by oxidation of the decyl moiety to several polar metabolites. A negligible amount of total radioactivity remained in the body after one week (Henderson, 1992).
- A closely related compound, didecyldimethylammonium iodide, was not well absorbed by the dermal exposure route (Henderson, 1992).
CHRONIC CLINICAL EFFECTS
- No treatment-related changes other than mild skin irritation were seen in rats exposed to DDAC subchronically at levels of 2, 6, and 12 mg/kg/day by the dermal route. There was virtually 100% mortality in mice given 3000 ppm DDAC in the diet for several days; deaths were from severe wasting and dehydration. The No Observed Effect Level (NOEL) was 600 ppm, corresponding to 107 to 134 mg/kg/day (Henderson, 1992).
- In a 90-day subchronic study rats, a dietary level of 3000 ppm produced 80% mortality with inflamed and fluid- or gas-filled intestines. A concentration of 1000 ppm or less produced no treatment-related effects (Henderson, 1992).
- Chronic oral doses of 3 to 20 mg/kg/day had no effect on mortality, organ weights, or gross pathology in dogs, but 20 mg/kg/day decreased erythrocyte counts, hemoglobin and hematocrit values. Increased incidences of excessive salivation, emesis, and soft/mucoid/liquid feces were seen at 10 or 20 mg/kg/day. The NOEL for systemic effects was 10 mg/kg/day (Henderson, 1992).
- Some quaternary ammonium compounds are sensitizers and can cause allergic contact dermatitis or asthma. Among these are the closely related compound, quaternium-15, which has been frequently linked with occupational allergic contact dermatitis (Tosti et al, 1990). Benzalkonium chloride has caused occupational asthma (Innocenti, 1978). DDAC has been reported not to be a sensitizer in either humans or guinea pigs in unpublished studies (Henderson, 1992).
